Azido Palmitic Acid is an azide-functionalized C16 saturated fatty acid designed for bioorthogonal click chemistry and lipid-based applications. The terminal azide group provides a versatile reactive handle for azide–alkyne cycloaddition, including copper-catalyzed azide–alkyne cycloaddition (CuAAC) and strain-promoted azide–alkyne cycloaddition (SPAAC) with strained alkynes such as DBCO and BCN.
The hydrophobic palmitic acid chain promotes association with lipid bilayers, liposomes, lipid nanoparticles (LNPs), and other hydrophobic environments, enabling incorporation of an azide-functionalized lipid into lipid assemblies. Following incorporation, the azide group can be used to conjugate fluorescent probes, targeting ligands, polymers, biomolecules, and other alkyne-functionalized compounds through efficient bioorthogonal click reactions.
