Description
Synonyms: thiol-PEG-NHS; thiol-x-N-hydroxysuccinimide
Thiol-X-NHS (HS-x-NHS) is one of CBP’s thiol-ene click chemistry reaction reagents that can be used to conjugate to proteins, peptides and other material surfaces bearing primary amine groups.
NHS (N-hydroxysuccinimide) is an organic compound with the formula (CH2CO)2NOH. The NHS ester end binds specifically to primary amines (-NH2), which are abundant in proteins and peptides. Reactions between NHS and an amine group results in a stable amide bond, a degradation-resistance linkage between two molecules in the cell. Meanwhile, thiol (-SH) reacts easily with enes such as maleimide or carbonylacrylics to form stable cyclic conjugates, as well as gold, silver, and other thiol-reactive surfaces. There is a medium length PEG linker between thiol and NHS groups that offers better water solubility and sufficient space between substrates and thiol groups to enable more specific and efficient binding.
Common applications of this molecule include surface functionalization by coating gold nanoparticles or biosensor chips, bioconjugation linking proteins to drug carriers, and hydrogel formation crosslinking polymers to create biocompatible scaffolds.
