Norbornene (NBE) DSPE is a phospholipid conjugate containing a norbornene-functionalized head group covalently attached to 1,2-distearoyl-sn-glycero-3-phosphoethanolamine (DSPE). The norbornene moiety enables rapid and selective inverse electron-demand Diels–Alder (IEDDA) click reactions with tetrazine-functionalized molecules under mild, catalyst-free conditions.

The hydrophobic DSPE lipid readily incorporates into liposomes, lipid nanoparticles (LNPs), and cell membranes, while the exposed norbornene group provides a convenient handle for post-assembly surface modification. This strategy enables efficient attachment of fluorophores, antibodies, polymers, nucleic acids, and other biomolecules without disrupting membrane integrity.

Norbornene DSPE is widely used in targeted drug delivery, membrane labeling, biomaterials research, and diagnostic imaging applications. Compared with trans-cyclooctene (TCO), norbornene offers improved chemical stability while maintaining excellent bioorthogonal reactivity toward tetrazines.
