Carbonylacrylic (CBA) Palmitic Acid is a CBA-functionalized C16 fatty acid conjugate designed for bioorthogonal click chemistry and lipid-based applications. The carbonylacrylic moiety provides a reactive handle for rapid and selective inverse electron-demand Diels–Alder (IEDDA) reactions with thiol molecules under mild, catalyst-free conditions.

The hydrophobic palmitic acid chain promotes association with lipid bilayers, liposomes, lipid nanoparticles (LNPs), and other hydrophobic environments, while the exposed carbonylacrylic group enables subsequent surface functionalization. This approach allows fluorescent probes, polymers, targeting ligands, and other CBA-functionalized molecules to be introduced through a convenient bioorthogonal conjugation strategy.
