DMPE Norbornene (NBE) Conjugate is a phospholipid functionalized with a norbornene group through 1,2-dimyristoyl-sn-glycero-3-phosphoethanolamine (DMPE). The norbornene functionality undergoes rapid and selective inverse electron-demand Diels–Alder (IEDDA) click reactions with tetrazine-containing molecules under mild, catalyst-free conditions, providing an efficient platform for bioorthogonal conjugation.

The DMPE lipid anchor contains two C14 myristoyl chains, enabling efficient incorporation into liposomes, lipid nanoparticles (LNPs), lipid bilayers, and other membrane-based systems. Compared with DSPE derivatives, the shorter acyl chains of DMPE exhibit reduced membrane retention and can provide greater lipid mobility or faster exchange in certain formulations, making DMPE conjugates well suited for applications requiring dynamic membrane modification.

Following incorporation into lipid assemblies, the exposed norbornene group serves as a reactive handle for site-specific attachment of fluorophores, antibodies, polymers, nucleic acids, and other biomolecules through bioorthogonal click chemistry. DMPE-NBE Conjugate is widely used in targeted drug delivery, membrane engineering, diagnostic imaging, and nanoparticle surface functionalization.
