Tetrazine DPPE is a phospholipid conjugate containing a norbornene-functionalized head group covalently attached to 1,2-dipalmitoyl-sn-glycero-3-phosphoethanolamine (DPPE). The tetrazine moiety enables rapid and selective inverse electron-demand Diels–Alder (IEDDA) click reactions with alkene- and alkyne-functionalized molecules under mild, catalyst-free conditions.
The hydrophobic DPPE lipid readily incorporates into liposomes, lipid nanoparticles (LNPs), and cell membranes, while the exposed carbonylacrylic group provides a convenient handle for post-assembly surface modification. Tetrazine DPPE is widely used in targeted drug delivery, membrane labeling, biomaterials research, and diagnostic imaging applications.
